The 12th fret symbolizes the crucifixion. It shows the two vines converging on the cross where Jesus defeats death. In the new Jerusalem, there is only the tree of life. The tree of the knowledge of good and evil, which causes death, is no more. The 12th fret on the banjo is the half way spot between the neck and the body. When a harmonic is played there, the strings don't move even though there is sound. That silence seems fitting when we consider the meaning of Jesus' crucifixion.
